All plasmids are maintained in transformed bacteria (in the -80˚C freezer) for long-term storage, sharing, and for easily making DNA preps when needed.
- When you have constructed a new plasmid, prepare a frozen stock and give it a systematic name, as explained in the "Making Bacteria and Plasmid Stocks" protocol in the lab BoxSync > Protocols > Common Protocols folder.
- Keep the frozen stock in the Pending box in the -80°C freezer for now.
- Print the Plasmid Notes form. Complete both pages, but leave the "Bacterial Stock Position" blank.
- Hand it to Emanuel. He will enter the information and assign a position in the Bacterial Stocks boxes in the -80°C.
- Transfer the frozen stock to the new position when ready.
